We have previously shown that Bone morphogenetic protein 7 (BMP-7) prevents and restores the loss of gut function associated with TNBS induced colitis. In an attempt to address the question of specificity of this action regarding other bone morphogenetic proteins we have examined the efect of of BMP-6, the closest BMP-7 "relative", on the gut function using the same experimental model.Yet, BMP-6 like BMP-7, has protected intestinal tissue from extensive damages in the course of an inflammatory reaction induced by TNBS, the overall effect of BMP-6 on the restoration of normal gut structure and function was less prominent. Furthermore, some clear distiction observed between BMP-6 and BMP-7 effects have undoubtedly confirmed specificity of the BMP-7 anti-inflamamtory properties which could be, at least in part, ascribed to the modification of Th1 mediated cellular response. Suppression of the IL-12 expression at the mRNA level observed upon BMP-7 therapy in the intestinal tissues from colitic rats was accompained by supression of the osteopontin (Eta-1) expression, a key cytokine for mounting an efficient Th1 immune responses.Interestingly, BMP-7 treatment did not influence the expression level of Bcl-2, Bax and p53, the main regulators of the cell cycle and cell death - the processes behind pathogenetic mechanisms of inflammatory bowel disease (IBD).
