Micropropagation of some Croatian endemic plant species
A rapid clonal propagation method for three Croatian endemic plant species: Fibigia triquetra (DC.) Boiss., Degenia velebitica (Deg.) Hay. and Centaurea ragusina L. has been developed. Shoots originated from aseptically germinated seeds collected on natural habitats were used for culture initiation. The highest multiplication rate in a 4-weeks-culture period was achieved on half strength MS medium supplemented with 2.9 mM gibberellic acid and 0.5 mM 6-benzylaminopurine (BA) in Fibigia triquetra (9.2 shoots per explant) and Degenia velebitica (5.2 shoots per explant) cultures, or with addition of 1.0 mM BA in Centaurea ragusina (4.6 shoots per explant) cultures. Excised shoots (2-4 cm) were rooted on the same basal medium with addition of 8.6 mM indole-3-butyric acid (IBA) for Fibigia triquetra shoots, 4,9 mM IBA for Degenia velebitica shoots and 2,5 mM IBA for Centaurea ragusina shoots. Rooted plantlets of all three species were successfully transferred to potting soil and acclimatized to outdoor conditions.
