Mapping broomrape resistance in faba bean

Broomrape (Orobanche crenata Forsk.) resistance in faba bean (Vicia faba L.) is a complex trait, difficult to manipulate in breeding programmes. Thus, the objective of our research is to identify genomic location and gene action of individual quantitative trait loci (QTLs) controlling broomrape resistance in order to develop a marker-assisted selection (MAS) programme. In order to develop a linkage map, a population consisting of 196 F2 plants was derived from the Vf6 x Vf136 cross. The maternal parent, Vf6, was highly susceptible to O. crenata, whereas Vf136 showed a fair level of resistance. F3 lines, obtained by selfing F2 individual plants, were studied for broomrape resistance under field conditions in order to identify and map QTLs associated with this trait. Recombinant inbred lines (F6 RILs) from this cross were developed in order to confirm the presence of the QTLs found on F2 linkage map as well as their stability in different environments. In order to analyse the accuracy of the map developed in the F2 generation, based predominantly on dominant markers we have compared it to the map derived from F6 generation. The 165 RI lines were evaluated for broomrape resistance in the field trials carried out in Córdoba in 2003 and 2004 and in Mengíbar in 2004. The linkage map developed in F2 mapping population consisted of 121 markers mapped into 16 linkage groups covering 1445.5 cM of the faba bean genome. More saturated linkage map has been developed in RILs consisting of 254 markers mapped into 19 linkage groups covering 2.811 cM. We were able to assign nine out of 19 linkage groups to their specific chromosomes. A high degree of correspondence between maps has been observed. Three putative QTLs (Oc1, Oc2, and Oc3) controlling broomrape resistance were identified in F2 population using composite interval mapping approach. Two out of these (Oc2 and Oc3) were also detected in RILs. Oc2, assigned to chromosome VI, explaining about 10% of the phenotypic variability of the trait was detected in Córdoba 2003 and Mengíbar 2004 trials. Oc3, located on chromosome II, was detected in Córdoba 2003 and 2004 trails explaining up to 22% of the phenotypic variation. Moreover, new genome regions on chromosome I conferring resistance has been detected. Further validation of putative QTLs across multiple test environments will be carried out as well as the saturation of the F6 map in genomic regions associated with broomrape resistance.
