Crowat project: Assessment of metallothioneins in gills of field-collected mussels Mytilus galloprovincialis as a biomarker of metal exposure

Mussels Mytilus galloprovincialis were collected at four coastal sites in the middle Dalmatia, Croatia (Kaštela and Trogir Bay). The metal and metallothionein (MT) analysis were performed in the heat-treated cytosol of gills. MTs were determined by differential pulse polarography, and metal concentrations by atomic absorption spectrometry. The gill mass and the condition index were determined as the indicators of biotic conditions. Statistical analysis of the data obtained at all four stations within four-year monitoring indicated to simultaneous increase of MT concentration and the total cytosolic protein concentration (TP) (r=0.57 ; p<0.001), and to dilution of both parameters with the increasing gill mass (MT: r=-0.23 ; p<0.1, TP: r=-0.51 ; p<0.001). Further on, MTs were normalized on TP concentration, and expressed as MT/TP ratio. According to the statistical analysis performed on the entire data set, following conclusions can be drawn: - positive influences of water temperature, gill mass and cytosolic Cu concentration on MT/protein ratio were obtained ; - cytosolic Zn concentration followed the changes of the TP level, both showing an opposite trend to MT/TP ratio ; - the sites with higher level of Cu usually had higher level of Zn, too, probably as a result of the specific anthropogenic activities. The results will be discussed in terms of suitability of gill MTs, isolated from field-collected mussels, as biomarker of metal exposure. Natural variability should be distinguished from contaminant induced stress, taking into account that sampling sites in Kaštela and Trogir Bay differ in biotic conditions, mussel size, age and specific contaminant sources.
