QTL mapping for ascochyta blight resistance in faba bean

Ascochyta blight is a common disease of faba bean (Vicia faba L.) caused by Ascochyta fabae Speg. In order to identify quantitative trait loci (QTLs) controlling faba bean resistance to A. fabae linkage maps were developed using both F2 and RIL (Recombinant Inbred Lines) populations derived from the cross between resistant (Vf6) and susceptible line (Vf136). F3 lines, obtained by selfing F2 individual plants as well as an entire RIL population were tested for ascochyta blight resistance after inoculation under growth chamber conditions. Disease severity was scored as the percentage of symptomatic area of the leaf. The linkage map developed in F2 mapping population covered 1445.5 cM of the faba bean genome. More saturated linkage map has been developed in RILs covering 2.811 cM. Using F2 mapping population two putative QTLs controlling ascochyta blight resistance (Af1 and Af2) were identified, located on chromosomes III and II, respectively. The same genomic regions were also detected in RILs. Fine mapping of these resistance genes will help in marker-assisted selection for ascochyta blight resistance in V. faba, facilitating gene transfer and pyramiding of the genes into acceptable genetic backgrounds.
