Database for PKSS and NRPSS: computer programs to automatically find and annotate new sequences

Recent DNA sequencing projects (e.g. genome sequences) are yielding a large number of new potential PKS and NRPS modular clusters whose functionality and products are unknown. A program has been written to scan the EMBL database using BLAST to identify potential clusters. This program is run regularly and used to identify new clusters that have been added to the EMBL database. The new clusters are downloaded and screened for potential modules using further programs. The output is then screened by eye to decide if a complete cluster is likely to be present. Further programs analyse these potential clusters to identify domains. By using models for the structure of typical modules it is possible in most cases to assemble the domains into modules. Operator intervention is necessary if atypical module structures occur. The final program enters the newly annotated clusters into the database. Clusters are only entered after a human decision that the annotation is appropriate. This system will allow the rapid addition of novel sequences that have not been adequately annotated previously. There will be a computer demonstration of programs.
