S6 haploinsufficiency is associated with activation of a p53-dependent checkpoint during gastrulation

Ribosome biogenesis is required for the large increase in the rate of protein synthesis that occurs during cell growth and proliferation. Even though ribosome biogenesis is intense during mouse oogenesis, this pool of ribosomes is not sufficient to support an enormous growth of the embryo during early embryogenesis. Therefore, the synthesis of new ribosomes is activated at the six- to eight-cell stage embryo. An error in ribosome biogenesis could have a deleterious consequence on oogenesis and embryonic development through impaired protein translation. Additionally, molecular mechanisms that sense the fidelity of ribosome biogenesis may exist. To gain insight into these processes we induced a defect in ribosome biogenesis by conditionally deleting one allele of the 40S ribosomal protein S6 gene in the growing mouse oocytes and generated S6-heterozygous embryos by appropriate breeding.
