Adjuvant activity of peptidoglycan monomer and structurally related afamantylpeptides-the effects on humoral immune response in mice
Peptidoglycan monomer*, GlcNAc-MurNAc-L-Ala-D-isoGLN-meso-DAP (wNH2)-D-Ala-D-Ala (PGM) originating from Brevibacterium divaricatum and synthetic adamantyltripeptides, isomers of D,L-(adamant-2-yl)-glycyl-L-Ala-D-isoGln (AdTP1 and AdTP2) exhibit immunomodulating activity (1,2). The purpose of the present work was to compare the effects of these compounds on humoral immune response in mice. BALB/c and C57Bl mice were immunized with ovalbumin (OVA). Control groups received OVA in saline (50 mg/mouse) and separate groups received respective tested compounds in addition to OVA, also in saline, in dose of 100 or 200 mg/mouse. Tested compounds were administered with the first injection only, or with first injection and boosters, in different experiments. Booster injections were given on days 14 or/and 28. Anit-OVA IgG was assayed by ELISA, developed for this purpose, in the sera taken from mice 7 days after the booster injection. IgG subtypes were determined by commercial Bio-Rad kit. IgG anti-OVA levels were significantly higher in all experiments in groups of mice treated with PGM with respect to controls and groups treated with AdTP1. AdTP2 treatment resulted in significantly higher increase in anti-OVA IgG after the first booster and with higher doses of AdTP2 also after the second booster. PGM and AdTP2 induced significantly higher levels of IgG1 and IgG2 anti-OVA subclasses than detected in controls and AdTP1 treated groups indicating that these immunomodulators might upregulate both Th1-like and Th2-like immune response. *Produced by PLIVA, Zagreb, Croatia 1 Tomašić et al.
