Effect of sialoadenectomy and lacrimal glands removal on already estabilished bpc abillity to promote corneal epithelial defects healing in rats

Purpose. We have shown in our previous experiment that BPC 157 promotes corneal epithelial defects healing in rats. In this experiment we investigate whether sialoadenectomy and lacrimal glands removal ( together with growth factors present in the glands ) would anulate the positive healing effect by BPC . Methods. albino Wistar rats (salivary and lacrimal glands removed), 200 g b.w. were randomly divided in groups. After intraperitoneal and topical anesthesia, circular lesions were induced to the both eyes by abrading corneal epithelium with surgical lancet under surgical microscope all the way to the limbus. The defects were stained with standard florescein and photographed. Medication distilled water (control group) or BPC 157 2pg/ml , 2 ng/ml, 2&micro ; ; g was started either immediately after injury induction every 8 h up to 40 hours or in the condition of already advanced lesion, 8h after injury induction and carried up to 40 hours. The healing process was monitored by photogarphy of fluorescein stained cornea before application .The defect area stained green was determined by morphometrical analysis using a sprecial program VAMSTECH. The size of defect area is expressed as a precentage of total corenal area. Then the data was compared between groups to see whether there was a difference in heeling size area Results. At the earliest assessment point, 8 h after injury, the lesions became smaller in all groups but there was no significant difference in healing size when compared to the controls. At later intervals differences was significant in both cotreatment and postreatment groups( mg, ng) when compared to controls. Conclusion. The failure of already established BPC (mg, ng) effect to promote defect healing at 8 h in cotreatment grops sugests that BPC closely colaborates with growth factors from lacrimal and posibly salivary glands in corneal healing during early phase of healing. Further uninterupted coarse (8-48) shows that BPC has a healing ability of its own throuout the remaining healing peroid. In post treatment groups , the BPC aplication 8 h after injury of oadenectomized rats with clear BPC healingg effect supports this assumption.
