Differential gene expression in callus and tumour tissue lines of sugar beet (Beta vulgaris L.).

Normal, habituated and transformed in vitro tissue lines of sugar beet (Beta vulgaris L.) were studied with regard to patterns of cellular, extracellular proteins and isoesterases. A correlation between developmental processes and protein patterns was established. Electrophoretic patterns of cellular proteins were rather similar for all tissue lines. Silver stained gradient polyacrylamide gels showed few specific bands. The polypeptides of 16 and 17 kDa were found in normal and tumour tissue, the one of 37 kDa was tumour specific. The bands of 18 and 19 kDa were higher expressed in unorganised habituated tissue than in other tissue lines and the polypeptide of 23 kDa was characteristic for organised habituated tissue. In extracellular protein patterns differences were, at least quantitatively, more obvious. The polypeptides of 33, 23 and 13.5 kDa were not detected in extracellular matrix of unorganised habituated tissue, which contained more protein bands of higher molecular mass (54 or more kDa). This line is characterised by lack of cellulose and lignin in cell walls and reduced cell-to-cell adhesion (Tacchini P. et al., Plant Physiol. Biochem. 1995, 33/3/:361). The majority of extracellular proteins were glycosilated except the ones with molecular mass below 22 kDa. After incubation of the gels in solution containing 1-naphtylacetate and 2-naphtylacetate as substrates the isoesterases were visualised showing distinct patterns for different tissue lines.
