Crystallisation and powder data collection of the lysozyme from acetate buffer at pH 3.5-9.6.
Lysozyme was first enzyme to have its three-dimensional structure determined by X-ray crystallography. Lysozyme can be easily crystallized in various crystal forms. It is of interest to investigate the possibility of structure refinement with powder X-ray diffraction data collected on the laboratory X-ray diffractometer. The sample was measured on an X'Pert PRO Multi-Purpose Diffractometer (MPD), configured in transmission geometry with a focusing mirror on the incident beam side and an X'Celerator detector on the diffracted beam side. The raw data were indexed by the DICVOL program integrated into the HighScore Plus software suite. Indexing results showed very good agreement with the data obtained from single crystal diffraction.
