Very little is known about the molecular mechanisms that integrate processes of cell growth and cell cycle progression in mammalian cells. Since ribosome biogenesis has been associated with cell growth and cell division, we considered the possibility that it coordinates these two events. To test this hypothesis, we introduced a defect in ribosome biogenesis by conditionally deleting one allele of the S6 ribosomal protein gene in the mouse embryos or T cells by using Zp3-CRE or CD4-CRE transgenic mice, respectively. Our results show that the deletion of one S6 allele prevented embryonic development at the onset of gastrulation and blocked proliferation of T cells upon engagement of antigen receptors. Furthermore, they strongly suggest that a p53-dependent checkpoint is activated by a change in the fidelity of the translational machinery in vivo to eliminate defective cells or prevent aberrant cell division. Such studies will be essential in elucidating the molecular mechanisms involved in coordination of cell growth and proliferation during the development and evolution of human disease states.
