Intraoperative photodynamic therapy successfully eliminates large tumor in mice. Background and purpose: Photodynamic therapy based on the light application on the tumor containing photoactive components, is limited by the depth of light penetration in the tissue. The longer the light wave length the deeper the penetration. Red light used in these experiments could penetrate 3 to 4 mm in the tissue, i.e. the tumor. Light penetration into the tumor could be increased either by opening the skin covering the tumor or by reducing tumor surgically. The results obtained could be important in clinical use of photodynamic therapy. Material and methods: CBA/HZgr mice and transplanted fibrosarcoma were used. Photofrin II, 2, 10, 15 and 20mg/kg, respectively and red light (250mW/cm2), were the components of photodynamic therapy. Results and conclusion: Photodynamic therapy was successful if higher doses of Photofrin were used. Further, tumor volume of 120 uL could be eliminated significantly more effective if the skin over the tumor was opened. If the tumor had reached the volume of 500 uL photodynamic therapy was without any benefits, i.e. none of treated tumors disappeared. However if such a big tumor, in Photofrin II treated mice, was surgically reduced immediately prior to the exposure to the light photodynamic treatment was effective, i.e. 5/7 tumorous mice were cured. Thus, combination ofsurgery and photodynamic therapy is effective in experimental model used and may present hope for clinical application.
