Changes of experimental allergic encephalomyelitis (EAE) in AO and DA rats by intracerebroventricular or subcutaneus application of somatostatin
The influnce of peripheral (subcutaneus) and central (intracerebroventricular)administration of somatostatin analogue Sandostatin (SMS 201-995)on EAE was analysed in two strains of rats: relatively resistant Albino Oxford (AO) and susceptible Dark August (DA). Application of different doses of SMS was made in early and late postimmunization period and the effects were evaluated by clinical score of disease, phenotypic analysis of cells in spleen and lymph nodes, functional reactivity of lymphocytes to antigen specific and unspecific challenge, and by immunohistological findings in CNS. The data revealed that the effects of both types of application are dependant on a dose and time of somatostatin administration.The greatest changes in DA rats were observed after SMS given on the seventh day after active immunisation, when the great mortality and prolongation of relapse was observed.Similar effects were noticed in resistant AO rats, in which SMS provoked the disease in 55% of rats, in comparison with only 17% in control, injected with saline. The time kinetic FACS analysis showed that relapse and recovery phase of EAE in SMS-treated rats induces the characteristic changes in the proportion of W3/25, OX8, OX-1 and OX-39+ cells in spleen and lymph nodes, as well as the variations in the intensity of blastic transformation to Con A.Before the relapse, the latter was significantly increased in comparison with saline treated group, indicating that SMS-induced changes in systemic and local immunity contribute to the altered susceptibility to EAE induction in both strains of rats.
