Immunomodulating effects of peptidoglycan monomer linked with zinc in old mice ;

It is well known that age-associated decline of immune functions are often connected with the zinc deficiency. In the experiments presented herewith we tested the effects of new immunomodulationg agent peptidoglycan monomer (PGM), in which the basic substance was linked with zinc (PGM-Zn). Its effects in short lived BALB/c mice, aged 10-12 months, were compared with the effects of equimolar doses of PGM and ZNCl2.The treatment lasted 26 days (one i.p. injection, every fifth day). The results have shown that PGM-Zn markedly enhances the antibody production to SRBC, as well as the spontaneous and Con-A-induced blastogenesis. Plaque forming cells generation in individual mouse was poositively and negatively correlated with the total quantity of hepatic proteins. PGM-Zn induced also the appearance of peritoneal macrophages which in cocultures with syngeneic splenocytes were less able to enhance the spontaneus, and particulary the Con A induced blastic transformation.Enhancing activities of PGM-Zn in some aspects more correlated with the action of PGM, whereas, the induction of suppressive macrophages more correlated with the activity of ZNCL2. The data emphasize that PGM-Zn may activate both, stimulating and inhibiting pathways in immunosenescence, by mechanism which are not identical with the activity of PGM or ZnCl2.
