Immunomdulating effects of peptidoglycan monomer linked with zinc (PGM-Zn) in mice depleted of CD4+ lymphocytes
We previously observed that PGM-Zn possesses the immunocorrective effects in some immunodeficient states. To obtain more information about the mode of its action, in this work we analyted its effects in mice, which were in vivo depleted of CD4+ cells.The analysis included the estimation of primary and secondary type of immune response to sheep red blood cells(SRBC), measurement of concentration of antigen specific and unspecific immunoglobulins in sera, determination of cellularity of lymphatic organs, phenotype of cells in spleen, phagocytic capacity of peritoneal macrophages and the estimation of class II MHC expression in the liver. The data revealed that in spite of the absence of CD4+ lymphocytes, PGM-Zn is able to stimulate the plaque forming cells production to SRBC in both primary and secondary reaction. In primary reaction this was followed by remarkable increase of antigen unspecific and specific IgM titer in sera, by hyperplasia of spleen and elevation of total number of CD4, CD8, CD5 and Ig positive cells. In secondary reaction the titer of IgG and IgG1 was also increased, but cellularity of spleen and its content of CD5 and Ig+ cells were decreased. In both reactions PGM-Zn increased the expression of class II expression in the liver and number of cells in timus and bone marow and stimulated the phagocytic activity of macrophages. The data suggest that immunocorrective potential of PGM-Zn on humoral immunity may be linked with its ability to upregulate class II expression on mononuclear phagocytic system and induce the polyclonal activation of B lymphocytes.
