Effect of organic selenium (Sel-Plex®) supplementation and a 48 h fast post- fattening on antioxidant enzymes activity and lipid peroxidation in tigh and breast muscle of broiler chickens
The effect of organic selenium (Se) supplementation on the activity of antioxidative enzymes and lipid peroxidation in tigh (M. biceps femoris) and breast (M. pectoralis mayor)muscle in chicken (ROSS 308) was investigated. At 42 days and after 48-h fast, 10 birds out of a group of 40 broilers fed a standard diet (control group, <0.15 ppm Se as sodium selenite) and 10 out of a group of 40 broilers fed the same diet topdressed with 0.3 ppm organic selenium as Sel-Plex® ; ; (Alltech Inc.) were killed. In the tigh and breast samples of both groups, the activity levels of glutathione peroxidase (GSH-Px), catalase, manganese superoxide dismutase (Mn-SOD) and tiobarbituric acid reactive substances (TBARS) were determined. data were analysed by ANOVA followed by a Duncan's post hoc test. In tigh muscle of broilers given Sel-Plex® ; ; , activity of GSH-Px was significantly higher (P<0.02)after the fattening period, and GSH-Px (P<0.001) and catalase activity (P<0.05) were significantly higher after the fasting period than in chickens fed with standard diet. At the same time, after the fast, activity of Mn-SOD was significantly lower (P<0.05) than in the control group. The fasting period resulted in a significant increase of Mn-SOD activity (P<0.0001) and TBARS (P<0.01)in thigh muscle of the control group, and in significant increases of Mn-SOD (P<0.01) and catalase (P<0.05)activity in tigh muscle of broilers supplemented with Sel-Plex® ; ; . In the breast muscle of control group, after the fasting period, a significant decline in GSH-Px (P<0.05)and Mn-SOD(P<0.05)activity and TBARS (P<0.0001)concentration were found. Simultaneously, in the breast muscle of chickens given Sel-Plex® ; ; the fasting period resulted in a significant decline in Mn-SOD activity (P<0.001)and TBARS concentration(P<0.0001), but no decline in GSH-Px activity was noted. It was concluded that after the fattening period, as well as during the fasting period, chickens supplemented with organic selenium (Sel-Plex® ; ; ) more effectively maintained antioxidative status via enhanced GSH-Px activity compared with birds fed the standard diet.
