Effects of indomethacin on polymorphonuclear leukocyte and monocyte phagocytosis and oxidative burst in vitro

Indomethacin has been one of the most actively investigated nonsteroidal antiinflammatory drugs (NSAIDs) and is believed to act mainly through mechanisms of cyclooxygenase inhibition and synthesis of prostaglandins. Its effects on phagocytosis, however, have not been extensively studied. In this report effects of 10-5M and 5x10-5M concentrations of indomethacin on polymorphonuclear granulocyte (PMN) and monocyte (Mo) ingestion, intracellular killing and oxidative burst in vitro were investigated in 10 healthy volunteers. Indomethacin at both concentrations significantly potentiated PMN and Mo intracellular killing. At a lower concentration it also stimulated PMN/Mo oxidative burst. The higher concentration stimulated PMN oxidative burst, although less than the lower one and was without effect on Mo oxidative burst. There was a highly positive correlation between PMN/Mo oxidative burst and intracellular killing at lower, oxidative burst-stimulating concentration of indomethacin. It was supposed that effects on PMN/Mo intacellular killing were realized mainly by enhancement of oxidative mechanisms due to downregulation of prostaglandin synthesis mediated by inhibition of two isoforms of cyclooxygenase. Stimulatory effects on PMN and Mo intracellular killing ability and oxidative burst were attained with indomethacin concentrations which could be achieved during antiinflammatory therapy.
