Shoot regeneration in petiole explants of Vitis vinifera L.
An attempt was made to establish a large-scale propagation method of the Dalmatian grapevine cv. Vugava by iduction of adventitious buds. Bud induction and shoot formation were obtained from the leaf petiole explants derived from microcloned shoots. Adventitious were induced after explant culturing on the basal medium containing mod. MS macronutrients, Heller's micronutrients, 2.5 uM 6-benzyladenine (BA) and 5 uM IBA. Shoots did not regenerate on the media with other ratios of BA(5.0, 1.0 uM) to IBA (0.1, 0.5 uM). Explants generated adventitious shoots directly from the explants wihout callus formation. After two months in culture the regenerated shoots were transferred to the medium with 0.2 uM IBA. Shoots rooted in three weeks and developed into entire plants.
