Influence of explant and gas exchange on shoot rejuvenation of cv. Vugava grapevine in vitro
The difference among phenotypes in culture of grapevine cv. Vugava was found depending on the degree of shoot rejuvenation in vitro. Shoots obtained in the initial culture by nodal segment cultivation on medium MS supplemented with 7.0 uM BA and 5.0 uM IAA were the most similar to the adult stage (by tendril number and distribution and distichous phyllotaxy). In the course of subculturing on the same medium or by shoot development from small vegetative tips, adult morphology gradually disappeared in favour of better pronounced juvenile phyllotaxy and rarefaction of tendrils. Plants with complete juvenile properties were reached from adventitious shoots regenerated in the leaf petiole segments on mod. MS macro-, Heller's micronutrients, supplementented with 2.5 uM and 5.0 uM BA, or by culturing mature embryos on Nitsch and Nitsch (1969) medium supplemented with 10.0 uM IAA and 1.0 uM GA3. In the test tubes wrapped by Parafilm shoot growth was slower, plant organs size was irregular and vitrification was observed. Shoots with pronounced juvenile properties were more suitable for rapid micropropagation. However, it should still be examined to what extent will they affect the plants of such origin.
