Factors affecting of grapevine cultivars, Vugava and Plavac mali micropropagation from in vitro cultured minicuttings

The micropropagation of grapevine cvs Vugava and Plavac mali throught nodal segment method was studied. For culture establishment green, one-node cutings from the vineyard plants were used. Murashinge and Skoog (MS) medium supplemented with 4.4 microM was the most useful to induce bud growth in buth cultivars. After two to four weeks of culture, axillary shoots were excised from nodal segments and transferred to the sama basal medium with the addition of 11.4 microM IAA for for further four weeks. This method was sutable for the culture establishment of both cultivars. Growing shoots were excised to obtain new one-node sterile minicuttings that were again cultured. The average number of nodes per shoot through seven successive subcultures was 4.4. Nodal segment method was suitable for the micropropagation of Vugava cultivar. 6-benzyladenine was more efficient in axillary bud induction in comparison with kinetin. In Vugava cultures optimum shoot and root growth was obtained on MS medium supplemented with 11.4 microM IBA ; the growth of axillary buds was faster in younger nodes of the donor plant. Nodal segment method has a potential value in commercial clonal propagation of Vugava cultivar. In the culture of Plavac mali, shoot growth was inconsistent and in vitro propagation was strongly limited.
