Thin Layer Chromatography in Textile Conservation

Thin layer chromatography is used to simultaneously separate many different components from their mixture, and to identify unknown materials by comparing their RF values to that of known reference materials. By applying modern instruments for automatic application of samples and automatic recording and evaluating of plates and by being simple, rapid, and relatively inexpensive, this method holds advantage over many alternative analytical techniques. Different substances which have been used through the history as binders on paintings, sculptures, wooden or textile materials, such are proteins, waxes, carbohydrates or resins, can be identified by this method. The identification of these materials is often crucial in order to plan future conservations treatment steps and to understand deterioration processes. For this reason it is important to characterize the technologies employed by artists. The main advantage for textile conservators is the possibility to identify many different binding substances (proteins, waxes, resins or polysaccharides) which can be present on textile materials. The aim of this work was to optimise the composition of a mobile phase for thin layer chromatographic separation and identification of binding media on historical textile. The experiment included ultrasonic extraction of the resin sample, hydrolysis of referent protein materials and binding media from the textile sample, separation of amino acids in hydrolysed sample by thin layer chromatography and densitometric determination of the components.
