Evaluation of Heteromorphism of DA/DAPI Bands Using Imaging Analyzer Software
Clinical significance of heteromorphism of constitutive heterochromatin is still obscure since its function remains unknown. The use of molecular technology allowed the discovery of many new variants to which authors mostly refer as normal heteromorphisms, without phenotypic consequences. Although the amount of heterochromatin varies considerably in human chromosomes 1, 9, 16 and Y, a total amount of constitutive heterochromatin in the cell tends to remain more or less constant. We tend to develop a simple technique for the objective evaluation and quantification of heterochromatin in the cell. For this purpose we used the imaging analyzer software (IPLab Spectrum, version 3.1) and a selective staining technique (DA/DAPI) for human metaphase chromosomes of 30 persons. Heteromorphism of chromosomes 1, 9, 16, Y and D-group chromosomes was evaluated by measurements of the area and fluorescence intensity. For both measurements, different transformations were used and compared.
