Tissue metal content and metallothionein I + II protein expression in mouse brain, lungs and kidney after exposure to gasoline vapor

Mammalian metallothioneins (MTs) are a group of low-molecular weight, cysteine rich proteins with high affinity for both essential and nonessential metals. In vivo, the metal-binding involves mainly Zn2+, Cu+, Cd2+ and Hg2+, while in vitro additional and diverse metals such as Ag+, Au+, Bi3+, Co2+, Fe2+, Pb2+, Pt2+, and Tc4+ may be bound to apothionein (the metal-free form), showing a role in metal ion homeostasis. However, as multipurpose proteins MTs also have important function in protein&#8211; protein and protein&#8211; nucleotide interactions participating in mitochondrial respiration, thermogenesis, energy metabolism, angiogenesis, as well as in cell survival and differentiation, having a role in cytoprotection against various types of injuries. Moreover, owing to their important neurophysiological and neuromodulatory functions MTs might be involved in a range of acute and chronic neurological disorders. In an attempt to elucidate some aspects of these events, in this study we investigated the effects of airborne pollutant-gasoline (G) vapor on tissue metal components and MT I+II expression in the brain, lungs and kidney. Experiments were done on C57/BL6 mice, which were for one h/day exposed to G vapor in a metabolic chamber. Control groups consisted of intact mice and of those exposed to the similar degree of restrain stress. Protocol was repeated for three or 10 days and then the expression of MT-I and MT-II and tissue content of Zn2, Cu2+ and Fe2+ were analyzed in the brain, lungs and kidneys, using the immunohistochemistry and inductively coupled plasma spectrometry and atomic absorption. The data showed that G inhalation markedly upregulated the MT expression in the brain, affecting ventricular ependyma, parenchymal astrocytes, cells lining the perivascular space and particularly the hippocampal glial cells in the subgranular zone of dentate gyrus, i.e. at the sites of neurogenic niches. High MT expression was also found in alveolar epithelia and in the kidney, implying the protective effects of MT in several areas exposed to toxic agents or their metabolic products. Simultaneously, in G-exposed mice significantly decreased concentration of Zn2+, Cu2+ and Fe2+ (p<0.01) in the brain and increased concentration of Cu2+ in the kidney (p<0.01) was found, suggesting that MTs through abstraction or donation of metals participate in reestablishment of morphostasis after injury.
