Primary cultures - a model for expression analysis of HH-GLI signaling pathway
We used primary cultures of ovarian dermoids as a model for study of the HH-GLI signaling pathway. Several genes involved in the pathway were analyzed by Real-Time PCR. We prepared primary cultures of ovarian dermoid tissue, isolated each colony that formed on the plate and cultured it separately. In that way we obtained several clone lines from each primary culture. Expression of PTCH, GLI1, SMO and SHH genes was determined in each clone line separately. Analysis of these clones showed differences in Hh-Gli pathway gene expression, and, therefore, proved the heterogeneity of these tumors. Blocking with cyclopamine and induction with Shh showed some of these lines are responsive to cyclopamine and Shh protein. These results show that cyclopamine treatment could be used for potential therapy of these tumors. We intend to check these effects on primary cell cultures of different tumors.
