Propolis and Honey Increase the Antimetastatic Ability of Chemotherapeutic Agents in Murine Tumor Models

We investigated the antimetastatic efficiency of water soluble derivative of propolis (WSDP) or honey given perorally (po) alone or in combination with chemotherapeutic agents. Transplantable murine tumors were used: a spontaneous mammary carcinoma (MCa) and a methylcholanthrene– induced fibrosarcoma (FS) of CBA mouse, and an anaplastic colon adenocarcinoma (ACa) of Y59 rat. Metastases in the lung were generated by injecting viable tumor cells intravenously (iv). WSDP exerted a significant antimetastatic effect (p < 0.05) when given either before or after tumor cell inoculation. In a combined treatment with WSDP and Epirubicin a profound inhibition of metastasis formation was found ; this synergistic effect is maximal when Epirubicin and WSDP were administrated after tumor cell inoculation. A pronounced antimetastatic effect (p < 0.05) was also achieved given alone or in combination of honey with 5-fluorouracil (5-FU) or Adiamycin (ADR) before tumor cell inoculation (2 g/kg for mice or 1 g/kg for rats, once a day for 7 consecutive days) as compared to control. However, honey given to animals after tumor cell inoculation enhanced metastastatic growth. Positive outcome of combined treatment with either propolis or honey and chemotherapeutics was expressed by the influence of these treatments on peripheral blood leukocytes. We have shown that a possible mechanism(s) of a combined treatment with chemotherapeutics and honey bee products could be the consequence of modulation of cytochrome P450 (CYP) enzymes included in biotransformation of chemotherapeutics. To conclude, these findings indicate that the combination of WSDP or honey with chemotherapeutics could increase the antimetastatic potential of chemotherapeutic agents and decrease their adverse effects such as leukopenia.
