Influence of apolipoprotein E gene on lipoprotein profile in patients with vascular dementia and in healthly octogenarians

Apolipoprotein E (apo E) is a protein associated with plasma lipoproteins. Apo E polymorphism has been related to significant modifications of lipoprotein profile, as well as to the incidence of different pathologies including vascular dementia and Alzheimer's disease. Furthermore, it was proposed that apo E polymorphism might be involved in the aging selection process. To evaluate Apo E polymorphism in patients with dementia and healthy octogenarians, impact of apo E polymorphism on plasma lipids and serum apo E concentrations in all groups of examiners. 148 Croatian subjects including 51 demented patients, 45 healthy octogenarians and 52 healthy adult controls were studied. Lipoprotein parameters (total cholesterol, triglycerides, HDL-cholesterol, LDL-cholesterol) and apoprotein E were measured by standardized methods. Apo E genotypes were determined by ARMS methods which includes PCR (polymerase chain reaction). The frequencies of e2, e3 and e4 alleles in patients with vascular dementia, were 0.07 ; 0.72, and 0.21 ; in elderly examines 0.08 ; 0.80 and 0.12 ; in control group 0.06 ; 0.84 and 0.95. Differences in lipoprotein parameters emerged, a trend toward higher total and LDL-cholesterol from the e2 to the e4 type was observed. In all study population the e4 allele had a raising effect, while e2 had the opposite effects on total cholesterol levels. The serum apo E levels decreases in e4 carriers. These results suggest that apoE gene is not significantly associated with longevity in Croatian population, but that e4 allele is still related with vascular dementi.
