Protective effect of sialic acid in fructose induced human lens protein modification

We investigated the effect of sialic acid, a keto hexose, on the formation of advanced glycation end product(AGE) in human lens protein, induced by in vitro glycation by fructose and glucose. Sialic acid inhibited fluorescence quenching by 15 percent. This probably reflect sialic acid transition metal chelation capacity, shown by 20 percent inhibition of Fe ferrozine formation. Sialic acid did not affect significantly neither the non-tryptophan novel fluorophore formation nor the tryptophan quenchinng induced by glucose. These results suggest that sialic acid exert its protective effect probably mainly through impeding fructose induced conformational changes of lens protein, and also in paert by its antioxidant action.
