Lipid monolayer structure of high density lipoproteins
The dynamic of the lipid monolayer of the native lipoproteins was monitored by ESR spectroscopy with nitroxide spin labels and by fluorescence spectroscopy with induced fluorescence probe. The empirical partition parameter for TEMPO and steady state anisotropy for TMA-DPH indicated a single behaviour lipid compartment, in spite of high protein/lipid ratio. The mobility and accessibility of the probes to the quenching reagents indicated lipids heads are more apart with less mobile tails in HDL3 than in HDL2. The restriction in lipid mobility induced by oxidative modification of the particle was measurable in HDL2 but only slightly detectable in HDL3 indicating that rigidity of the smaller particle structure has minor changes upon oxidation while on the other hand the restriction of lipid mobility in HDL2 indicates the ordering of the lipid tales and diminishing of their wobbling motions. Based on our results we siggested that the lipid mobility restriction is highly dependent to protein/lipid ratio and that enviromental induced changes in more rigid HDL3 have less influence on the structure of the particle than in bigger and more fluid HDL2. We argued it might be the course of the difference in the antiatherogenetic behaviour of the HDLs.
