Haplotype analysis of 17 Y-STR loci in southern Croatian population

To define the Y-chromosome genetic structure in Southern Croatia population. The blood samples were collected at the Department of Forensic Medicine, University Hospital Split, from unrelated healthy male. Average age of participants was 40 and their all known ancestors were of Southern Croatian origin. All individuals gave their informed consent together with the approval of the authorized Ethics Committee. DNA was extracted from peripheral blood by the Chelex method. Y-STR haplotypes were determined from a sample of 166 men from Southern Croatia using the AmpFlSTR Yfiler PCR amplification kit (Applied Biosystems) that includes the 9-loci European minHt, the 11-loci SWGDAM core set (minHt plus DYS438 and DYS439) and, in addition, the markers DYS437, DYS448, DYS456, DYS458, DYS635 (Y GATA C4) and Y GATA H4. Electrophoresis and typing were performed in the ABI PRISM 310 sequencer. Allelic frequencies were estimated by using gene counting method. Haplotype diversity within population was calculated by using the Arlequin 2000 package The 17-loci Y-STR haplotypes, allele frequencies and locus diversity of 166 individuals are shown in Table 1 and Table 2. There were 152 different haplotypes observed. Total haplotype diversity was 0.997289, and 141 haplotypes (84.49 %) appeared as unique. Haplotype No.27 was the most common haplotype and shared by 4 males in the sample. The range of locus diversity was between 0.21292 in DYS392 to 0.75546 in DYS439 locus. Combination of these 17 STR loci presents a powerful tool for individual identification and parentage analysis in the Southern Croatian population.
