The influence of fat free diet on plasmalogens in brain
Fat free diets (FFD), because of the lack of essential fatty acids, can cause some changes in the content and the composition of lipids. The aim of our study was to elucidate the effects of fat free diet on fatty acid composition in rat brain plasmalogens. Animals were fed FFD, isocaloric with the control diet (CD), for two weeks, and were sacrificed by bleeding. Total lipids were extracted by solvent mixture (CHCl3:CH3OH) and plasmalogens were separated by two-dimensional HPTLC. The composition of sn-1 alkenyl radicals and sn-2 fatty acids for plasmalogen-phosphatidylethanolamine (PPE) fraction was determined by gas chromatography. By comparing the results obtained for CD and FFD, the following effects have been revealed: (a) an increase in C16 fatty acid desaturation; (b) a decrease in arachidonic acid content, followed by an increase in eicosapentaenoic acid synthesis, and (c) no significant changes in sn-1 alkenyl radical composition were found. According to these results it can be concluded that this type of diet, relative to plasmalogens, supports the stabilisation of membrane permeability.
