Abstract Background and purpose: Bee propolis and its compounds are the most promising extract as antitumor agents. Previous research has confirmed that ethanolic extract of propolis (EEP) and its compounds exhibited antitumor and antimetastatic properties, due to immune stimulation, free radical scavenging, alteration of the mitotic cycle of tumor cells, anti-angiogenesis activity, apoptosis induction, or combination of these effects. Up to now, no study has demonstrated a reduction of solid tumor growth with water soluble derivative of propolis (WSDP), and there is no research concerning the antitumor effect of caffeic acid. This study examines the effect of polyphenolic compounds (caffeic acid-CA, caffeic acid phenethyl ester-CAPE) deriving from propolis and WSDP itself on the tumor growth in vivo and in vitro. Material and methods: A transplantable mammary carcinoma (MCa) of CBA mouse and culture of human cervical carcinoma cells (HeLa) were used. Tumors were generated by 1x105 or 1x104 viable tumor cells given subcutaneously (sc). Test compounds were given intraperitoneally (ip) or per os (po) at doses of 50 or 150 mg/kg before or after tumor cell inoculation. Results: The results indicated that WSDP and its polyphenolic compounds reduced the tumor volume and increased life span when given prophilacticaly ; however given after tumor cell inoculation were less effective in this respect. In in vitro studies all test components but WSDP clearly inhibited the growth of HeLa cells. Conclusions: These findings indicated that WSDP and CA reduced tumor growth when given to mice before inoculation of tumor cells ; these results corresponded well with prolongation of life span in treated animals.
