Analyses of IgG1 and IgG2a Immune Responses Specific for Ovalbumin in Mice adjuvanted with novel PGM-containing Montanide ISA formulations
We have recently developed novel oil-based adjuvant formulations (ISA720PGM and ISA206PGM) by incorporation of immunostimulating peptidoglycan monomer (PGM) into Montanide ISA720 and ISA206 oil-based adjuvants1. So far they have shown excellent adjuvanticity in improving humoral immune response in rabbits to several tested antigens. Here we wanted to investigate the influence of novel formulations on Th1/Th2-type of cellular immune response with respect to the induction of IgG isotypes. CBA (H-2k) and NIH/OlaHsd (H-2q) mice were immunised s.c. with ovalbumin (OVA) as antigen adjuvanted in ISA720 and ISA720PGM or ISA206 and ISA206PGM respectively. Positive control group was immunised with OVA formulated in CFA/IFA. IgG as well as IgG1 and IgG2a as specific indicators of Th2 and Th1 type of immune responses respectively, were determined by ELISA after the first and the second booster. For evaluation of Th1/Th2 type of immune response the ratio of IgG1 to IgG2a (IgG1/IgG2a) was calculated for each serum in experimental groups and compared to the ratio of no adjuvant-treated group. The IgG1/IgG2a of no adjuvant-treated group indicated Th2-biased immune response specific for OVA. Both Montanide ISA adjuvants shifted immune response towards Th1, with ISA720 being significantly stronger of the two and also very similar to CFA. The influence of PGM incorporation didn't seem to influence the established immune response direction.
