Perforin expression is increased in the lesional epidermis of patients with severe and mild psoriasis
Background: The significance of cell-mediated cytotoxicity in psoriasis is still not well understood. Cytotoxic T lymphocytes and NK cells mediate cytotoxicity reactions mainly by perforin/granzyme and Fas/FasL pathway. Objective: Perforin expression and distribution of various lymphocytes subsets in psoriatic lesional and non-lesional skin were studied in patients with severe and mild disease. Methods: Skin biopsy specimens from lesional and non-lesional skin of 9 patients with severe, 8 patients with mild disease and 8 healthy persons were analyzed by immunohistochemistry. Results: We found a significant increase in epidermal CD4+ and CD8+ T cells in lesions of both, severe and mild disease. Perforin expression was significantly enhanced in the lesional epidermis of patients with severe and mild disease. Conclusion: Perforin expression is up-regulated in the lesional epidermis of patients with severe and mild disease suggesting a role of this cytolytic mechanism in the creation of psoriatic lesions in both, patients with severe and mild disease.
