Crystal and molecular structure of the bromo insulin derivative at room temperature
Insulin is a hormone that regulates the glucose level in blood (carbohydrate metabolism) and also takes part in metabolism of fat and proteins. Insulin is used medically in patients with Type 1 diabetes mellitus. Owing to its important biochemical role and pharmaceutical importance insulin is one of the most extensively researched molecules. The insulin molecule consists of two chains (A and B) linked by two disulfide bonds. In the presence of Zn ions and some other divalent cations (Co, Ni, Mo, Cu) insulin forms hexamers. There are three forms of insulin hexamers: T6, T3R3 and R6 forms 1 . These forms of insulin are used in therapeutic preparations for the control of diabetes. Here we report the structure of the bromo derivative of human insulin at room temperature. Single crystals were grown by the hanging drop vapour diffusion crystallization method. Diffraction data sets up to 2.71 Å ; ; resolution range were collected on the laboratory diffractometer Oxford Diffraction Xcalibur 3 with MoKα radiation. Reduction was performed by the CrysAlis RED program. Unit cell-parameters are a = 80.96 Å ; ; and c = 37.30 Å ; ; . The c axis was found to be halved in comparison to the bromo insulin derivative structure at 100 K 2 . The bromo insulin derivative belongs to the T3R3f hexamer. Coordination of zinc ions and conformation of the insulin molecule will be discussed.
