Biological Agents targeting Interleukin-18
IL-18, initially cloned as an interferon (IFN)-γ inducing factor, is an important regulator of both innate and acquired immune responses. It is up-regulated in several human autoimmune and inflammatory diseases, and, therefore, might represent a novel therapeutic target. This review highlights the biology of IL-18, its central role in inflammation and immune response, and the various therapeutic strategies of IL-18 blockade, including recombinant human IL-18BP, anti-IL-18 monoclonal antibodies, soluble heterodimeric IL-18R, and the inhibition of IL-18 secretion via inhibition of caspase-1 or antagonism of the P2X-7 receptor.
