Attenuation of lipid peroxidation during mice liver regeneration by polyphenols extracted from olive oil

Liver regeneration is accompanied by increased level of lipid peroxidation (LPO), resulting from excessive oxygen free radicals (ROS) production, that can be modified by dietary antioxidants. Antioxidant properties of polyphenols originating from the olive oil are well established and are partially attributed to their ability to scavenge ROS. Although beneficial effects of the olive oil on the liver regeneration have been already recognized, little is known about the contribution of the polyphenols to this process. Accordingly, the effect of the polyphenols extracted from the olive oil on the hepatic lipid oxidative damage after 1/3 partial hepatectomy (pHx) and the implication on the liver regeneration was studied in mice. For that purpose C57/BL6 mice were assigned in four groups: PFE-mice treated with polyphenols extract, pHx-mice subjected to 1/3 partial hepatectomy, PF+pHx-mice treated with polyphenols extract followed by partial hepatectomy, controls-mice receiving vehicle alone. A dose of 50 mg/kg bwt of the polyphenols extract was intraperitoneally injected daily on 7 successive days. Mice were sacrificed 24 h after pHx and the liver was removed. LPO was established by measuring malondialdehyde (MDA) in liver homogenates using thiobarbituric acid reaction. A calculated relative liver weight was used to assess the liver regeneraton ratio. As expected, pHx induced marked increase in liver MDA level with respect to the controls. Although the level of MDA in PFE+pHx group was higher than in the control group, treatment with PFE prior to pHx significantly reduced liver MDA level when compared to pHx animals. Furthermore, the relative liver weight in the PFE+pHx group was significantly higher than in the pHx group. Consequently, pre-treatment with olive oil polyphenols enhances regeneration ratio of remnant liver tissue, presumably due to their antioxidant properties, as indicated by attenuated LPO level.
