Application of 16S ribosomal RNA sequence in the identification of laboratory strains of Bacillus species in the warfare polymer fibre

The microbiological analysis showed that, in the both cases, in the polymer fibre Bacillus brevis bacterium. However, based on the analysis of 16S ribosomal RNA sequence, two bacterial species were detected in the polymer fibre: Bacillus megaterium MPF 90 with 1503 base pairs (Figure 2) and Bacillus cereus G8639 with 1502 base pairs per a sequence analysed. The resistance on synthetic antibiotics – sulfonamides (sulfamethoxazole, trimethoprim) indicated that these strains should be considered as „ laboratory strains“ . The test on the presence of plasmid DNA showed that the bacteria do not have plasmid, and that means that the genes for resistance to antibiotics are on the chromosomal DNA. The results of the research showed that the polymer fibre is an important military tactic system that might be used as a carrier of different pathogenic microorganisms or chemical warfare agents. It was established that the polymer fibre is a complex system made of 3 components: nylon, gelatine substance (mixture of polysaccharide and protein), and bacteria placed inside. It is proved that the bacteria have certain pathogenic characteristics that cause inflammatory reactions in the organism. The identification of the bacteria was done in two ways: first by classic microbiological kites and second by the method of the identification through the comparison of the sequence 16S rRNA ; after that was examined the resistance to antibiotics and the presence of plasmids in a cell.
