Do antigen presenting cells help in removing tumor - associated glycoprotein -72 (tag-72) from the maternal-fetal implantation site?
Tumor - Associated Glycoprotein -72 (TAG-72 ) is a mucin that is expressed in a variety of human malignancies, as well as in the uterus, oviduct and ovary during the normal menstrual cycle. Weather TAG-72 is present in human decidua during the pregnancy is largely unknown. In this study we evaluated the expression of TAG-72 in the early human pregnancy mucosa, at the normal and pathological maternal-fetal interfaces by single or double immunohistology of paraffin embedded tissue sections. The internalizations of TAG-72 in normal decidual CD1a+ and CD14+ cells from isolated decidual momonuclear cells (DMC) were additionaly tested. Negative immunostaining of TAG-72 was seen in decidual tissue of normal first trimestar pregnancy decidua, blighted ovum and missed abortion. As a positive control ovarian tumor and mammary carcinomas were used. Localized TAG-72 espression was found in tubal mucosal surface and glandular epithelial cells close to the implantation site and away from the implantation site, but not in uterine mucosa of ectopic pregnancy. Double labeling revealed that some CD14 labeled cells close to the tubal implantation site contained TAG-72 positive granulas in the cytoplasm, whereas we were not able to detect TAG-72 in rare CD1a+ cells by serial sections labeling. Our data sugests that TAG-72 is down-regulated in uterine implantation sites in normal and pathological conditions, whereas it remained in traces in tubal mucosa close and away from the implantation site. Since TAG-72 decreased internalization of FITC-dextran by normal early pregnancy decidual CD1a+ and CD14+ cells, double positive tubal CD14+TAG-72+ cells might take part in removing TAG-72 from ectopic implantation site.
