Kinetic characterisation of enzymatic esterification in a solvent system : adsorptive control of water with molecular sieve
The kinetics of lipase catalyzed esterification of oleic acid and glycerol was studied in a batch stirred-tank reactor (BSTR). A commercial immobilized lipase (Lipozym) was used as the biocatalyst in solvent system with molecular sieve for water adsorption. The reactions were followed determining the reaction conversion during 45 hrs. Following the reaction in an organic solvent system with hexane as a solvent eliminates diffusion effects. Water formed during the reaction was removed by adsorption with molecular sieve, which was added at start of the reaction into the reaction mixture. The reaction rate constants was determined by solving the differential rate equations of the reaction system. The numerical values of the parameters were evaluated by fitting the model to the experimental data with the AURORA computer program, which performed the non-linear regression analysis using the Nelder-Mead method. Second-order irreversible kinetics model was found appropriate.
