Identifying protein flexibility by NMA
Here proposed apriori method for identification of protein flexibility relies on one end-state conformation of the protein. This method has comparative advantage over many others because it requires only one end-state conformation to efficiently identify protein flexibility. The background of this method relies on normal mode analysis as well as on several approximations and enhancements proposed in this work. Some of the enhancements are the development of enhanced anisotropic network model, integration of loopback procedure into clustering algorithm etc. These enhancements and some approximations have significantly decreased program complexity thus increasing computational speed. However, due to the possible accuracy drawbacks this method has been thoroughly tested. Accuracy testing of the method was conducted by comparing calculated and experimental temperature factors. Tests had shown significant correlation between experimental and calculated data. This comparison has been most useful to prove the accuracy of the results obtained by apriori method.
