Development of kinetic model for Trametes versicolor growth in submerged cultivation

Fungi fermentation is a complex process. Obtaining uniform pellets of a desired size is not easy, since many factors influence pellet formation such as inoculum size, type and age, media composition, shear forces, pressure, temperature, medium viscosity and oxygen concentration. Size and number of pellets are regulated by growth and breakage mechanisms. Trametes versicolor is white-rot fungi, known as a producer of lignolitic enzymes: lignin- peroxidase, mangenese-peroxidase and laccase. Although there is a huge interest for industrial application of this fungus and its enzymes, there is not many literature data about the kinetic of the growth of Trametes versicolor and enzyme production. In this work, a mathematical model of the process of pellet’ s growth and substrate consumption, during the batch cultivation, was given. For the description of the pellet’ s production, a three substrate Monod model for the biomass growth was given. Model parameters were estimated from the experimental data with different initial glucose and sucrose concentrations and for different initial pellets concentration. Model is validated and shows good correlation for wide initial concentration ranges of glucose, sucrose and biomass. Distribution of pellets growth during the fungus cultivation was also given, using image analysis technique.
