Flavonoids supplementation decreases oxidative DNA damage in lymphocytes in alloxan-induced diabetic mice

The present study was designed to investigate the possible antidiabetic and protective effect of flavonoids/polyphenols (quercetin, naringin, naringenin, chysin, caffeic acid, curcumin) against alloxan-induced oxidative DNA-damage in diabetic mice. Uptake of phytochemicals present in plants with especially high content of antioxidant flavonoids/polyphenols, might reduce oxidative cell damage. Type 1 diabetes was induced in mice by injection alloxan in dose of 75 mg/kg bwt iv. After 9 days of alloxan injection, there were an apparent reduction in the animal body weight and significant increase in serum glucose, triglyceride and total cholesterol as compared to the control normal group. On the other hand, daily treatment of animals with flavonoids in dose of 50 mg/kg bwt for period of 7 days ameliorated alloxan-induced alternations in the animal body weight as well as in serum glucose, triglyceride and total cholesterol. In addition, blood samples were collected after treatment of mice with propolis and flavonoids to analyze DNA damage (standard and modified comet assays). Alloxan produced slightly DNA single strand breaks as observed by increase in the % DNA in tail and tail moments of the comet. When the mice were treated with flavonoids, the DNA damage caused by alloxan was slightly reduced. In conclusion, polyphenols/flavonoids offer promising antidiabetic and hypolipidemic effects that may be mainly attributed to its antioxidant potential.
