Adamantyl-tenocyclidines as potential antidotes in soman poisoning
Continuing our work on investigation of various compounds (pyridinium, imidazolium and quinuclidinium derivatives) as potential antidotes in organophosphate poisoning we tested the therapeutic and protective effect of thienyl phenocyclidine (tenocyclidine, TCP) and its adamantylamine derivatives with piperidine (TAPIP), pyrolidine (TAPIR) and morpholine (TAMORF), respecitvely. All tested compounds were applied to the soman poisoned mice at dose of 2 mg/kg b.w. together with atropine (10 mg/kg b.w.). The therapeutic factors were in the range from 1.58 to 2.7, and therapeutic doses (all animals survived) from 1.25 to 2.0 LD 50 of soman. The best therapeutic effect was with TAPIP and TAPIR. The effects of tested compounds was higher when given 15 min. before soman poisoning. The same compounds were not very effective in tabun poisoning. All new synthesises compounds were characterised by m.p. determination, elemental analysis, and spectroscopic methods (IR, 1 H-NMR, 13 C-NMR, MS). In our experiments they were used as hydrochloride salts.
