Production and optimisation of microbial lipase

Industrial enzymes can be extracted from a naturally occurring source by production from a microorganism in fermentation. Lipases (3.1.1.3) belong to the class of serine hydrolases that break down triglycerides to fatty acids and glycerol. The fungus Aspergillus niger has been known to produce triacylglyccerol lipase. The extracellular lipase was produced by separating the cells from the fermentation broth without agitation in temperature controlled enviroment. Various parameters like initial pH, incubation period, effect of various nitrogen and carbon sources were optimised. Following the lipase activity, the best physicochemical conditions were found. It was observod that 72 hrs incubation, pH 6.0 and 30°C were optimum for growth, and production of lipase. The Monod equation was used for controlling the supply of a nutrient and the effect of some carbon and nitrogen sources were found. In all cases, the maximal specific rate of growth, u max, was determined. Besides the prediction of activities thorough computer programming was performed.
