Antifungal and antioxidative activity of some natural polyphenols
During invasion of yeasts into host, for efficiently disturbing hosts’ defences, several attributes must be satisfied. From one side, immunosuppression and damage of the natural defences of the host (physical, chemical) favour invasion of yeasts. Fungi also secrete putative virulence factors or demonstrate physical advantages (like polymorphism) to avoid with these strategies defence of the host. Damage of cells during fungal invasion cause a release of cytotoxic free-radicals. The aim of study was to compare antifungal activity and antioxidative capacity of some natural polyphenols: gallic acid (GA), tannic acid (TA), caffeic acid (CA) and epigallocatechin gallate (EGCG). The fluorescent dye exclusion method was used for detection of viability (apoptosis/necrosis rate) of Candida albicans ATCC 10231 together with inhibition of germination of blastospores in synthetic NAC medium. For analysis of antioxidative (scavenging of DPPH) capacity of polyphenols electron paramagnetic resonance (EPR) spectroscopy was used. Nystatin was used as control. Among polyphenols tested, EGCG and TA showed the highest rate of inhibition of germination of C. albicans blastospores (< 1.25 mg/mL) while other tested polyphenols were found to be active at 2.5 mg/mL. At 12.5 mg/mL all polyphenols significantly decrease of viability of blastospores with domination of apoptosis. The free-radical scavenging capacity (T50) of polyphenols was found to be about 30 s for GA, 110-130 s for TA and EGCG and 170 for CA while nystatin was inactive. Analysis of antifungal and antioxidative activities between natural polyphenols tested showed promising pharmaceutical application. The correlation between chemical structure and activity of polyphenols were explained.
