Short-term preservation of pink dentex (Dentex gibbosus, Rafinesque 1810) milt
The milt quality of pink dentex and possibility of the short-term preservation were examined. Milt was sampled from the live caught males by the abdominal massage method. Each sperm sample was split into two aliquots. The first sample was placed into the oxygenated plastic bag and the second sample was placed in bag containing ambient atmosphere. The samples were maintained at temperature of 4 °C. Milt volume and sperm concentration were measured. The range of spermatozoa concentration was 10.34-20.79 109/ml. The spermatozoa number was inversely proportional to milt volume and milt volume grows proportionally with the length of analyzed fish. The rate of sperm motility was estimated using 50 mM NaCl solution as the activating solution. After 62 h, sperm stored under oxygenated atmosphere had a significantly (P>0.05) greater percentage of motile spermatozoa (88%) compared to sperm stored under the ambient atmosphere (53.0%). After 9 days sperm stored under oxygenated atmosphere showed 50% motility and 5% after 11 days. These results suggest the importance of oxygen to the motility of pink dentex sperm and possibility of the artificial insemination. It makes significant step in future research on potential species for the mariculture production.
