Perforin mediated natural killer cytotoxicity
Cytotoxic lymphocytes are endowed with potent cytolytic machinery located in the cytoplasmic granules and containing cytolytic molecule perforin (pore forming protein) and associated granule proteases (granzymes). Perforin and granzymes are responsible for enforced death of noncooperative targets, whereas Fas and Fas ligand are responsible for induced suicide of compliant cells (perforin independent surface-receptor mediated pathway). Enforced cell death is critical for the elimination of parasitised cells. In unstimulated PBL perforin mRNA and protein is constitutively expressed in NK (high level of expression) and in the subpopulation of CD8+ cells expressing CD11b antigen (moderate expression). However perforin can be induced in CD8+CD11b- subpopulation and in some CD4+ lymphocytes. Perforin is constitutively expressed also in CD3+CD4-CD8- gama/deltaTCR subpopulation of PBL, but can be found also in CD3+CD4-CD8- alphabetaTCR+ subpopulation. Normal noninfected tissues are almost completely free of perforin+ cells. However, during pregnancy perforin+ cells appear abundantly in the granulated metrial gland (GMG) cells of the pregnancy in quantities higher than in any pathological condition.
