Human decidualized endometrial T lymphocytes express reduced levels of cytoplasmic CD3zeta

T lymphocytes comprise 5-10% of leukocytes in the human decidualized endometrium in pregnancy. The immunobiological function of these cells is unknown, but their T cell antigen receptor (TCR) has been reported to be downregulated in pregnancy. We have analyzed expression of cell cytoplasmic CD3zeta, a component of the TCR complex involved in intracellular signalling, in human first trimester decidual T cells using flow cytometric analysis of permeabilized cells. A mean of 75% of decidual T cells co-expressed cytoplasmic CD3zeta and cell surface CD3eta, which was not different from peripheral blood T cells (p<0.1). However, the levels of CD3zeta expression were significantly lower in decidual than peripheral T cells, assessed by mean fluorescence intensity (4.2 vs. 5.5 ; p<0.05), in the presence of higher levels of cell surface CD3eta (10.1 vs. 6.6 ; p<0.05). CD3zeta was not systemically downregulated since cytoplasmic CD3zeta levels did not differ significantly in peripheral blood T cells from pregnant women compared with non-pregnant controls (p<0.2). CD8+ T cells outnumber CD4+ T cells in decidua but the proportions of these two T cell subsets that expressed cytoplasmic CD3zeta were not significantly different. These results demonstrate that human decidual T cells locally downregulate CD3zeta, which may compromise their capacity for activation.
