Modulatory effects of SMS 201-995 on anti-CD3-induced apoptosis in thymus
It is already known that in vivo administration of anti-CD3 antibody to mice induces apoptosis in their immature thymocytes, predominantly in the double-positive (CD4+CD8+), CD3low cortical and CD3intermediate medullar cell subset. This activation-induced cell depletion is analogous to Ag-induced negative selection of thymocytes capable of recognising self antigens. Negative selection process is mediated by TCR and CD3 engagement, presumably via TCR-MHC complex interactions, and is additionally regulated through different intercellular contacts and humoral factors. Since the expression of somatostatin gene mRNA in thymus epithelial cells was found, in the present study we investigated the influence of somatostatin analogue SMS 201-995 (octreotide, sandostatin) on the susceptibility to apoptosis induction by anti-CD3 in immature thymocytes of BALB/c mice. Phenotypic analysis of DP thymocytes and nucleic fragmentation in thymus of mice injected with SMS and anti-CD3 antibodies were estimated at 24, 48 and 72 hours, as well as 5 and 15 days after treatment by using the cytofluorometric method on the flow cytometer. The data have shown that after the treatment with SMS, the anti CD3-induced decrease of DP-cells is less expressed. Additionally, SMS reduced the percentage of nucleic particles compared with a group of animals treated with antiCD3 alone, confirming that SMS may have an antiapoptotic effect. The data point to the possibility that somatostatin participates in the processes of T cell development and differentiation, and in those involved in clonal deletion of self reactive cells.
